Resumé:
Cryo-electron tomography (cryo-ET) allows obtaining the structure of macromolecular complexes in their cellular environment but not yet their full dynamics, due to the lack of appropriate image processing methods. The aim of this PhD thesis is to develop such cryo-ET image processing methods and use them to decipher in vitro and in situ pathways of conformational changes of the human ATPase p97, an essential molecular protein involved in several cellular processes (ERAD, membrane organisation, cell cycle regulation, etc.) and target for treatment of cancer and neurodegenerative diseases such as multisystem proteinopathy. This PhD thesis will be co-supervised by Dr. Jonic, CNRS-SU (image processing) and Dr. Rouiller, the University of Melbourne (structural biology of the ATPase p97), and will involve a stay of up to 1 year at the University of Melbourne. The methods and software that will be developed in this thesis shall also be useful for cryo-ET studies of other biomolecular complexes.
